Abstract
Osmoprimiranje je tehnika kojom se sjeme različitih biljnih vrsta djelomično ili u potpunosti hidrira s vodom ili pak otopinama različitih osmotskih vrijednosti s ciljem intenziviranja fizioloških procesa, kako bi se pospješilo i ubrzalo klijanje. U našem istraživanju je sjeme krastavca (Cucumis sativus L.) iz tri proizvodne godine, osmoprimirano s 300 μM otopinama dva različita donora sumporovodika: sporootpušajući donor GYY4137 i brzootpuštajući donor NaHS. Nakon imbibicije sjeme je osušeno i naklijavano na filter papiru navlaženom s 25 mM NaCl, odnosno vodom. Osnovna hipoteza je bila da H2S utječe na klijavost i pokazatelje vigora klijanaca kod sjemena krastavca naklijavanog u prisustvu soli te da postoje značajne razlike između primijenjenih donora H2S; NaHS i GYY4137. Pri osmoprimiranju s NaHS, najstarije sjeme iz 2009./2010. je imalo značajno nižu standardnu klijavost i energiju klijanja dok u varijanti osmoprimiranja s GYY4137 nisu utvrđene značajne razlike između godina deklaracije sjemena u navedena dva parametra. Klijanci dobiveni iz sjemena proizvodne godine 2010./2011. su u prosjeku imali najmanje mase hipokotila, korijena i kotiledona ali i najmanju prosječnu masu sjemena. Međutim, kod sjemena iz navedene godine deklaracije, tretman s GYY4137 je rezultirao značajno većom masom hipokotila u usporedbi s varijantom osmoprimiranja s NaHS. Također je utvrđen stimulativni učinak otopine NaCl na masu hipokotila u prosjeku za sve godine deklaracije i pri obje varijante osmoprimiranja. Dobiveni rezultati upućuju da osmoprimiranje s GYY4137 može povećati klijavost sjemena te akumulaciju svježe tvari u hipokotilima klijanaca razvijenih iz sjemena manje mase.

Osmopriming is a technique in which the seed of different plant species is partially or fully hydrated with water or solutions with different osmotic potentials aiming to intensify physiological processes, what improves and excellerates germination. In our study cucumber seed (Cucumis sativus L.) from three different production years was osmoprimed with 300 μM solutions of two different hydrogen sulphide donors: slowly releasing donor GYY4137 and fast releasing donor NaHS. After imbibition, seed was dried and germinated on filter paper moistened with 25 mM NaCl solution or water, respectively. Main hypothesis was that H2S influences germination and vigour of cucumber seedlings germinated in the presence of salt, and that applied H2S donors (NaHS and GYY4137) will differ significantly in their effect. In treatment with NaHS the oldest seed from production year 2009/2010 had significantly lower standard germination and germination energy, while in treatment with GYY4137 differences among seed production years in aforementioned parameters were not established. Seedlings germinated from seed produced in 2010/2011, had the lowest mass of hypocotyls, roots and cotyledons but also the lowest seed mass. However, seed from this year of production treated with GYY4137, developed significantly higher hypocotyl mass as compared to the treatment with NaHS. Also, stimulative effect of NaCl solution on mass of hypocotyls was seen regarding all years of seed production as well as in both osmopriming variants. The obtained results suggest that osmopriming with GYY4137 can increase seed germination rate and accumulation of fresh mater in hypocotyls of seedlings developed from smaller seed.
