Abstract
Osmoprimiranje je tehnika kojom se djelomično ili u potpunosti sjeme hidrira s vodom, različitim osmotskim otopinama ili solima, što potiče metaboličku aktivnost, povećava vigor i pospješuje klijanje. U ovom istraživanju, sjeme krastavca (Cucumis sativus L.) iz tri proizvodne godine (2009./10., 2010./11. i 2011./12.) je nakon osmoprimiranja donorima sumporovodika (H2S) različitih kemijskih svojstava, sintetskim sporootpuštajućim donorom GYY4137 te brzootpuštajućim donorom NaHS, naklijavano pri 25 mM NaCl odnosno vodi. Osnovna hipoteza je da H2S utječe na klijavost i pokazatelje vigora klijanaca kod sjemena krastavca naklijavanog u prisustvu soli te da postoje značajne razlike između primijenjenih donora H2S; NaHS i GYY4137. U prosjeku, klijanci uzgojeni iz mlađeg sjemena (2010./11. i 2011./12.) su imali značajno veću energiju klijanja te manje mase hipokotila i korijena u odnosu na klijance dobivene iz starijeg sjemena, najvjerojatnije zbog veće početne mase sjemena. Najstarije sjeme krastavca iz proizvodne godine 2009./2010. je imalo najnižu standardnu klijavost kod obje varijante osmoprimiranja sjemena donorima H2S što ukazuje na podjednaki efekt H2S, bez obzira iz kojeg je donora generiran. U varijanti naklijavanja u prisustvu soli je također utvrđena pojačana akumulacija svježe tvari, što upućuje na stimulativni učinak niske razine solnog stresa na masu hipokotila. Pošto povećanje masa hipokotila i korijena nakon primjenjenih tretmana upućuje na interakciju H2S i blagog solnog stresa, daljnja istraživanja je potrebno usmjeriti u razjašnjenje njihove pojedinačne uloge u fazi klijanja sjemena krastavca.

Osmopriming is a technique where the seed is partially or fully hydrated with water, different osmotic solutions or salts, which stimulates metabolic activity, increases vigor and promotes germination. In this study, cucumber seeds (Cucumis sativus L.) from three production years (2009/10, 2010/11 and 2011/12) has been germinated in the presence of 25 mM of NaCl or water, after osmopriming with hydrogen sulfide (H2S) donors having different chemical properties - synthetic slow releasing donor GYY4137 and fast releasing donor NaHS. The main hypothesis is that H2S influences on germination and seedlings vigor indicators of cucumber seed germinated in the presence of salt, and that there are significant differences in the effects of the applied H2S donors. In average, seedlings grown from younger seed (harvested in 2010/11 and 2011/12) had significantly higher germination energy and lower hypocotyls and roots weight as compared to seedlings obtained from older seed, most likely due a greater initial seed weight. The oldest cucumber seed, harvested in the season 2009/10, had the lowest standard germination rate for both variants of seed osmopriming with H2S donors, which points to the same effect of H2S regardless from which donor it has been generated. In a variant of germination in the presence of salt, the enhanced increment of fresh weight was also established, suggesting that low levels of salt stress may have stimulatory effect on the hypocotyl weight. Since an increase in hypocotyl and root weight after applied treatments suggests the interaction of H2S and mild salt stress, further research should be focused on clarifying their respective roles in germination stage of cucumber seed.
