Abstract
Cilj ovog istraživanja bio je utvrditi varijabilnost Rht gena u 20 hrvatskih i 10 stranih ozimih pšenica pomoću mikrosatelitnog markera gwm261 i specifičnih markera za Rht-B1 i Rht-D1 gene, te rezultate usporediti s visinom pšenice u poljskim pokusima. Identificirane su četiri različite alelne varijante na Xgwm261 lokusu, od 165, 174, 192 i 197 bp. Najzastupljeniji je bio alel od 192 bp, u određenim slučajevima dijagnostičnan za Rht8 gen, prisutan u 22 sorte (73,3%), odnosno 17 (85%) hrvatskih sorti. Ispitivanjem varijabilnosti na Rht-B1 i Rht-D1 lokusima, utvrđena je zastupljenost Rht-B1b i Rht-D1b alela kod 19 sorata (63,3%) odnosno 21 sorte (70%). Ovi aleli bili su prisutni u 80% hrvatskih sorata. Kombinaciju alela od 192 bp i Rht-B1b i/ili Rht-D1b alela imalo je 63,3% sorata. Gotovo sve sorte pšenice imale su očekivane visine s obzirom na prisutnost određenih alela na Xgwm261 lokusu, između 73,4 cm (Srpanjka) i 136,3 cm (U1), iako to nije bio slučaj kod prisutnosti različitih alela Rht-B1 i Rht-D1 lokusa.

The aim of this study was to determine the variability of Rht genes in 20 Croatian and 10 foreign winter wheat varieties using microsatellite marker gwm261 and specific markers for Rht-B1 and Rht-D1 genes, and to compare the results with wheat heights obtained from field trial. Four different allels were determined on Xgwm261 locus, 165, 174, 192 i 197 bp. 192 bp allel, in certain cases diagnostic for Rht8 gene, was predominant in 22 varieties (73.3%) and 17 Croatian varieties (85%), respectively. Analysis of variability on Rht-B1 and Rht-D1 loci showed presence of Rht-B1b and Rht-D1b allels in 19 varieties (63.3%) and 21 varieties (70%), respectively. These allels were present in 80% of Croatian wheat varieties. Combination of Xgwm261- 192 bp allel, and one or both Rht-B1b, Rht-D1b allel was present in 63.3% varieties. Wheat height varied from 73.4 cm (Srpanjka) to 136.3 cm (U1), and in most cases was as expected considering allel distribution on Xgwm261 locus, which was not the case for allel distribution on Rht-B1 and Rht-D1 loci.
