Abstract
Macrophomina phaseolina uzročnik je suhe ili ugljenaste truleži. U istraživanju su se koristili izolati Macrophomina phaseolina podrijetlom sa suncokreta i mrkve. Za istraživanje porasta micelija Macrophomina phaseolina korištene su četiri podloge; podloge od rajčice, mrkve, PDA i Čapek, a porast se pratio na četiri temperature 15, 22, 25 i 30°C. Za porast micelija Macrophomina phaseolina najpogodnije temperature su 25 i 30°C. Na tim temperaturama (kod oba izolata) micelij se razvio do ruba Petrijeve zdjelice (9,00 cm) prilikom trećeg i petog mjerenja, odnosno šesti i deseti dan od inokulacije. Na temperaturi 15°C micelij se vrlo slabo razvijao, te ova temperatura nije pogodna za razvoj. Najbolja podloga za razvoj micelija bez obzira na izolat na svim temperaturama je PDA podloga. Na 15°C kod izolata sa mrkve i izolata sa suncokreta mikrosklerocije su se razvile samo na podlogama PDA i podlozi od rajčice. Na temperaturama 22, 25 i 30°C najveće mikrosklerocije (bez obzira na izolat) izmjerene su na podlogama PDA i Čapek.

Macrophomina phaseolina is a pathogen that causes dry or charcoal rot. The used isolates were originated from sunflowers and carrots. Four growth media, tomatoes, carrots, PDA and Czapek and four temperatures, 15°C, 22°C, 25°C and 30 °C was used to study mycelia growth of Macrophomina phaseolina. The best temperatures for growth of the isolates were 25°C and 30°C. At these temperatures, both isolates mycelium has grown to the edge of the Petri dish (9,00 cm) in the third and fifth measurements, respectively sixth and tenth day of inoculation. At temperature of 15°C mycelium had poor development, thus 15°C is not suitable for development. The best medium regardless of isolates, for mycelium growth at all tested temperatures is PDA medium. At 15°C, isolates from carrots and sunflowers developed microsclerotia only on PDA and tomato medium. At temperatures of 22, 25 and 30° C the largest microsclerotia (regardless of isolates) were measured on PDA and Czapek medium.
